PCR技术在丙型肝炎病毒RNA聚合酶基因分析中的应用

摘    要:目的研究本地区HCV病毒分离株的分型,为临床治疗提供依据。方法利用PCR技术克隆并测定NS5B基因片段的核苷酸序列,并对其核苷酸和氨基酸序列进行比较分析。结果克隆的NS5B片段的核苷酸和氨基酸序列与HCV-Ⅱ型代表株相应部分序列的同源性最高,而且氨基酸序列比较结果显示,37个分离株中RNA聚合酶结构及其相关序列无任何变异。结论该分离株应属HCV—Ⅱ型。NS5B基因片段可能在病毒复制中起重要作用。

Abstract:Objective and Methods By using PCR technology we cloned and determined the nucleotide sequence in the gene fragments of NC5B,and its nucleotide and amino acid sequences were compared. Results The results showed that the cloned NS5B fragments of nucleotide and amino acid sequence nearly have the same character with the corresponding partial sequence in the HVC-Ⅱ type which indicating that should be HVC-Ⅱ type. Conclusion The result showed that the seven isolated viruses of RNA polymerase structure and its related sequences remained the same without any mutations, hence we suggested this research could play an important role in viral replication.
