胶质细胞源性神经营养因子基因转染雪旺氏细胞的实验研究

摘    要:目的应用胶质细胞源性神经营养因子(GDNF)基因转染雪旺氏细胞,观察GDNF基因在雪旺氏细胞内的表达效果,以及对雪旺氏细胞的营养作用。方法将雪旺氏细胞分为空白对照组、载体组、空载体组,通过阳离子脂质体介导,将GDNF基因转染雪旺氏细胞。用细胞增殖实验(MTT)、免疫组化、流式细胞仪观察在24h、48h、72h各组转染后对细胞的营养作用。结果在载体组细胞的生长状况较其他两组增殖良好。免疫组化可见GDNF在载体组细胞内呈现高表达。流式细胞仪观察基因转染后对雪旺氏细胞有明显的营养作用。结论GDNF基因转染雪旺氏细胞后,在细胞内呈现高表达,并对细胞有明显的营养作用。
