| 摘 要： | 目的探索无单克隆抗体亲和层析的重组人干扰素α2a分离纯化工艺。方法用 7mmol·L-1盐酸胍抽提干扰素 ,然后用金属螯合层析、离子交换层析、反相柱层析和凝胶过滤将提取的干扰素纯化。结果目标蛋白纯化了 195 1倍 ,蛋白比活性达 2 4 0× 10 8IU·mg-1,回收率达 30 1%。SDS PAGE检定呈一条带 ,其分子质量约为 19kD ,纯度大于 97%。结论本工艺降低了生产成本 ,易于扩大生产规模 ,适合产业化要求。 |

| Abstract: | Objective To research the isolation and purification process of interferon α2a without using monocloning antibody affinity chromatography.Methods Extracting interferon α2a using about 7 mol·L -1 guanidine hydrochloride and then purifying the extracted interferon α2a using metal chelate chromatography,ion exchange chromatography,RP HPLC and gel filtration chromatography.Results The target protein was purified by 195 1 folds.Its specific activity reached 2 40 × 10 8 IU·mg -1 protein.The yield was 30 1%.The purified product exhibited a single band on SDS PAGE and its molecular weight was about 19 kD .Conclusion This purification process at a low cost is easy to be scaled up and meet the requirements of production. |
