摘    要:目的:探讨丹参对体外培养大鼠成纤维样滑膜细胞增殖及 bax、bcl-2表达的影响。方法取浓度为1.5×10-5个/ml的 RSC-364成纤维样滑膜细胞悬液100μl加入细胞培养板,培养24 h,加入含不同浓度丹参的培养液,采用 MTT法分析丹参对成纤维样滑膜细胞增殖抑制作用及其时效关系;免疫组化法检测 bcl-2、bax表达情况。结果丹参可明显抑制大鼠成纤维样滑膜细胞增殖,并显示出一定的量效和时效关系。丹参可显著上调大鼠成纤维样滑膜细胞 bax表达,下调 bcl-2表达,与空白对照组比较差异有统计学意义( P 〈0.05)。结论丹参对体外培养大鼠成纤维样滑膜细胞的增殖具有抑制效应,其作用机制可能与调控 bax、bcl-2表达有关。

Effect of Salvia Miltiorrhiza on Fibroblastic Synovial Cells of bax/bcl-2 Expressions of Culture in Rats in Vitro

Abstract:Objective To explore the effect of Salvia Miltiorrhiza on fibroblastic synovial cells of bax/bcl-2 ex-pressions and proliferation culture in rats in vitro. Methods A concentration of 1. 5 × 10 -5/ ml RSC-364 fibroblastic synovial cells of 100 ul cell suspension was added with the cell culture plate,and was cultivated for 24 h,and then differ-ent concentrations of Salvia Miltiorrhiza culture medium were added. Inhibition effect of Salvia Miltiorrhiza on fibroblastic synovial cell proliferation and its time-effect relationship were analyzed using MTT method;bcl-2 and bax expressions were detected using immunohistochemical method. Results Salvia Miltiorrhiza was able to inhibit the cell proliferation of fibroblastic synovial cells in rats,and it showed a certain degree of concentration-response and time-effect relationships. Salvia Miltiorrhiza could significantly increase the bax expression of fibroblastic synovial cells,and reduce bcl-2 expres-sion of synovial cells,and the differences,compared with those in control group,were statistically significant( P 〈0. 05). Conclusion Salvia Miltiorrhiza can inhibit the proliferation of fibroblastic synovial cell culture in rats in vitro , and its mechanism of action may relate to regulation of bax and bcl-2 expressions.
