大鼠肠外营养模型的研究

摘    要:大鼠肠外营养模型的质量直接决定研究结果的可靠性。我科营养与代谢研究实验室自1988年建立大鼠肠外营养模型以来,不断地完善设备,提高手术技巧,重视无菌术并加强术后管理,建立了高质量大鼠肠外营养模型。在输液系统中采用金属施转轴,它可耐受高温消毒,具有良好的重复使用性,克服了套针性施转轴易漏液,不能耐受高温消毒,重复使用性较差等缺点。采用注射器式微电脑控制泵以提高输液的精确性和连续性。经颈外静脉作中心静脉插管,再经背扣,保护弹簧,施转轴与注射器式输液泵连接。输液速变控制在0.90ml·hr/100g鼠重手术时间20~30分钟。每日输氮1.62g/kg,非蛋白热卡1067.6KJ/kg,糖热卡占80%,脂肪热卡占20%。营养液在层流洁净台内配制。术后经过对周围环境反复清洁,消毒,控制室内温度和湿度等措施,使大鼠模型处在一个相对清洁的环境里。采用上述方法建立大鼠肠外营养模型可获得足够的热卡和氮量,体重有所增加,肝脏无明显病变。14天全部存活。

Abstract:The quality of parenteral nutrition model could affect the reliability of research results. The high quality parenteral nutrition model was established in our nutrition and metabolism Lab after improving the instruments, operative technique and intensifing the postoperative management.We used a metal swivel which could be sterilized in high temperature and be well reused, but the old swivel which was made of syringe was either easy to leak or poor reusable and could not be sterelized in high temperature.In order to infuse precisely and constantly, Computerized injectional pump was used to infuse nutritional liquid. The centre vein catheterzation was made through external jugular vein under aseptic operation and the catheter was connected to the injectional pump by a system composed by button, protective spring and swivel. The infusion rate was controlled in 0. 09ml/hr/100g body weight and the period of operation kept between 20-30 minutes. The nutrition formula was: Nitrogen 1. 62g/kg BW, Non-protein calorie: 1067. 6kJ/kg BW; in which the 80% was offered by glucose and anoter 20% by lipid. The nutritional so-lusion should be made up in the clean laminal hatch.The postoperative management should be well strengthed after applying such methods as cleaning, disinfeeting of the surroundings and controling the room temperature and humidity. The rats parenteral nutrition model could be set into a relatively clean surrounding.With the above methods, The rats parenteral nutrition model set up presently by our laboratory can get appropriate calories and nitrogen, along with increas in body weight and no obvious pathological change in liver. There is a possibility to survive for 14 days.
