羟苯磺酸钙对大鼠早期糖尿病肾病的保护作用

摘    要:目的:探讨羟苯磺酸钙对大鼠早期糖尿病肾病的保护作用及可能机制。方法:用链脲佐菌素(STZ)建立1型糖尿病大鼠模型。随机将24只成模大鼠均分为糖尿病模型组(D组)和羟苯磺酸钙治疗组(T组),另设正常组(N组)。于第4、8周末比较各组大鼠肾重/体重、24 h尿蛋白、尿液足细胞的排泄数目、肾小球正切面足细胞数目以及肾脏光镜电镜变化。结果:D组大鼠肾重/体重、24 h尿蛋白、尿液足细胞的排泄数目明显高于N组(P<0.05),肾小球正切面足细胞数目明显低于N组(P<0.05),T组大鼠肾重/体重、24 h尿蛋白、尿液足细胞的排泄较D组明显减少(P<0.05),肾小球横切面足细胞数目高于D组(P<0.05),病理变化也较D组明显减轻。结论:羟苯磺酸钙可以减轻早期糖尿病肾病大鼠蛋白尿而起到肾脏保护作用,其机制可能是减轻足细胞足突融合,减少足细胞的脱落。

Abstract:Objective: To investigate the protective effects of calcium dobesilate on renal injury in rats with early diabetic nephropathy.Methods: Rats injected with streptozotocin(STZ) were randomly divided into diabetic model group(D) and group(T) treated with calcium dobesitate.Another 12 normal rats were assigned into the control group(N).The ratio of kidney to body weight,24 h(urinary) protein,urinary podocytes excretion and podocyte numbers in the glomerular sections were analyzed after four and eight weeks.The renal morphology were also observed under light microscopy and electron microscopy.Results: The ratio of kidney to body weight,24 h(urinary) protein and urinary podocytes excretion were significantly higher in group D compared with those in group N respectively(all P<0.05),and glomerular podocyte numbers were significantly less in group D than in group N(P<0.05).The ratio of kidney to body weight,24 h urinary protein and(urinary) podocytes excretion were significantly lower in group T than in group D respectively(all P<(0.05).) Meanwhile,glomerular podocyte numbers were significantly less in group T than in group D(P<0.05).Renal pathologic changes were improved in group T.(Conclusion:) Calcium dobesilate ameliorates renal injury in diabetic rats through the protection effects of podocyte.
