三尖杉酯碱与阿糖胞苷联合诱导人白血病细胞分化的研究

小剂量三尖杉醋碱(LD-Harr)与小剂量阿糖胞苷(LD-Ara-c)联合治疗ANLL患者10例。在治疗期间,末梢血液和骨髓中原始细胞减少或消失时,伴有短暂的成熟中性粒细胞增多,且在其中见有Auer小体。新鲜分离ANLL患者的骨髓白血病细胞,加Harr和Ara-c进行液体培养后,白血病细胞分化为成熟的粒细胞,在其中也见有Auer小体。治疗结果,10例中有8例完全缓解,1例部分缓解,1例未缓解、副作用较轻。

Induction of Differentiation of Human Leukemia Cells by Harringtonine Combined With Cytarabine

Low dose harringtonine (LD-Harr) combined with low dose cytarabine (LD-Ara-c) was used in treatment of 10 patients with ANLL. In 2 of them a transient increase of mature granulocytes was noted in the peripheral blood and bone marrow when the blast cells disappeared from peripheral blood and decreased in bone marrow, concomitantly accompanied by the elevation of peripheral blood leucocyte alkaaline phosphatase activity, nitroblue-tetrazolium reaction and phagocytosis of zymosan particles progresively. During the combination therapy, Auer's bodies persisted in the cytoplasm of metamyelocytes on bone maJrow smear and in the. segmental neutrophils on blood smear in one of the patients. Freshly isolated leukemic cells from bone marrow of 4 patients with untreated ANLL were cultured in liquid in the presence of harringtonine combined with cytarabine. Apparent evidence of differentiation induction of leukemic cells and Auer's bodies in the cytoplasm of mature granulocytic cells was observed in two pationts. In the clinical study of 10 cases treated with combination of LD-Harr and LD-Ara-c, eight achieved complete remission and one partial remission. Prior to CR, mild degree of transient suppression of normal hemopoiesis was observed. The side-effects were acceptable and moderate.
