糖肾颗粒对大鼠肾小球系膜细胞增殖的影响

摘    要:[目的]研究糖肾颗粒对高糖培养下大鼠肾小球系膜细胞(Glomerular mesangial cells,GMC)增殖的影响,探讨其在糖尿病肾病(diabetic nephropathy,DN)防治中的意义.[方法]将常规体外培养的GMC分为正常对照组、高糖组、糖肾颗粒高、中、低剂量组及贝那普利组,分别进行相应处理后收集细胞,采用四甲基偶氮唑蓝(MTT)比色法检测不同作用时间(24h、48h、72h)下细胞增殖程度.[结果]高糖(30.0mmol· L-1)能明显刺激体外培养的GMC增殖.糖肾颗粒及贝那普利两种药物含药血清对高糖刺激下的GMC增殖有抑制作用,其中糖肾颗粒血清对GMC的抑制作用优于贝那普利血清,且呈一定的时间、剂量依赖性关系.[结论]高糖可促进GMC增殖,糖肾颗粒能明显抑制高糖条件下GMC增殖,从而发挥预防肾小球纤维化及硬化的发生、发展,进而延缓DN的进展.

Abstract:[Objective] To investgate the effect of Tangshen Particle on proliferation of rat glomerular mesangial cells cultivated with high glucose medium,and its significances on diabetic nephropathy prevention and control.[Methods] The rat glomerular mesangial cells cultivated with high glucose in vitro were divided into six groups: the control group,the glucose group,the Tangshen Particle high,medium and low dose groups and Benazapril group.Cells were collected after respectively given the appropriate treatment.The rat glomerular mesangial cells proliferative degree with in 24,48 and 72 hours were detected by MTT colorimetry[.Results] In vitro cultured glomerular mesangial cells were obviously proliferated under the stimulation of high glucose(30.0mmol/L).Both the serum of Tangshen Particle and Benazapril had significant inhibition on proliferation of rat glomerular mesangial cells,with the former better than the latter,and certain time and dose dependence were detected.[Conclusion] Tangshen Particle can inhibit GMC proliferation promoted by high glucose,which is helpful for the prevention of the progression of diabetic nephropathy.
