骨髓基质细胞输注对外周血干细胞移植造血恢复的影响

摘    要:为观察骨髓基质细胞输注对外周血干细胞移植(PBSCT)后造血恢复的影响,经放、化疗预处理的BALB/c小鼠,分别输入动员的外周血干细胞(PBSCT组)、外周血干细胞联合扩增的骨髓基质细胞(实验组),观察受体鼠4周的生存率、骨髓有核细胞(BMNC),粒细胞巨噬细胞集落形成单位(CFU-GM)、成纤维细胞集落形成单位(CFU-F)、外周血白细胞计数等指标。结果显示,实验组的生态率,BMNC、CFU-GM、CFU-F较PBSCT组显著升高(P<0.005);白细胞的最低值较PBSCT组高(P<0.01),白细胞恢复较PBSCT组块(P<0.01)。提示骨髓基质细胞输注有促进外周血干细胞移植造血恢复的作用。

Abstract:The present experiment was to study the effect of stromal cell infusion on hematopoietic recovery after peripheral blood stem cell transplantation in mice. BALB/c mice pre conditioned with high dose chemotherapy/radiotherapy were infused with peripheral blood stem cells (PBSCT group) or a combination of peripheral blood stem cells and bone marrow stromal cells (experimental group). Four week survival rate, white blood cell (WBC) count, bone marrow nucleated cells (BMNC), granulocyte macrophage colony forming units (GM CFU), fibroblast colony forming units (F CFU) were determined. The results showed that survival rate, BMNC, GM CFU, and F CFU were significantly higher in experimental group than that in PBSCT group ( P <0 05). Compared with the controls, the lowest WBC counts were significantly higher ( P <0 01), together with more rapid WBC recovery in the PBSCT group ( P <0 01). The results suggest that stromal cell infusion could enhance hematopoietic recovery after peripheral blood stem cell transplantation.
