芹菜素对小鼠睾丸标志酶活性的影响

摘    要:目的观察不同剂量芹菜素对小鼠睾丸标志酶活性的影响,探讨芹菜素对雄性小鼠生殖功能影响机制。方法成年健康SPF级昆明种雄性小鼠200只按体重随机分三个芹菜素剂量组(高1008mg/kg、中504mg/kg、低252mg/kg)和对照组(生理盐水),每组50只,每天定时灌胃,于7d、14d、21d、28d、35d后处死小鼠,取睾丸,制匀浆;比色法测乳酸脱氢酶(LDH)、酸性磷酸酶(ACP)、碱性磷酸酶(AKP)、葡萄糖-6-磷酸脱氢酶(G-6-PD)、烟酰胺腺嘌呤二核苷酸磷酸(NADPH)、琥珀酸脱氢酶(SDH)、一氧化氮合酶(NOS)吸光值并计算活力。结果LDH活力21d各剂量组高于同期对照(P〈0.05);ACP活力7d中剂量组高于同期对照(P〈0.05)、高剂量组高于同期对照(P〈0.01);G-6-PD浓度14d中、高剂量组高于对照(P〈0.01),35d低剂量组高于对照(P〈0.01)。AKP活力7d高剂量组低于对照(P〈0.05)。结论本实验条件下,芹菜素可弓}起小鼠睾丸组织酶活力的改变。

Study on effects of apigenin on activity of testicular marker enzymes in mice

Abstract:Objective To observe the effects of apigenin to testicular marker enzymes activity at different dose in mice, in order to explore the mechanism of the effects of apigenin on mice's reproductive system functions. Methods According to the body weight, the experimental groups were given different dose apigenin (high - dose group of 1 008mg/kg, medium -dose group of 504 mg/kg, low- dose group of 252 mg/kg), normal sa- line group ( 10ml/kg), 50 mice of each group were given corresponding handle with intragastric administra- tion at a certain time every day. Batch mice were killed in every 7 days, then got the testis. The light absorp- tion value of lactate dehydrogenase (LDH), acid phosphatase (ACP), alkaline phosphatase (AKP), glucose - 6 - phosphate dehydrogenase ( G - 6 - PD), nicotinamide adenine dinucleotide phosphate ( NADPH ), succinate dehydrogenase (SDH) and nitricoxide synthase (NOS) were detected by colorimetric method, and the vitality was calculated. Results The activity of LDH in three experimental groups were higher than that of the normal saline group at 21 - day (P 〈 0. 05 ) . The activity of ACP in medium - dose group was significant- ly higher than that of the normal saline group at 7 - day ( P 〈 0. 01 ) . The concentration of G - 6 - PD in the high -dose group and medium - dose were higher than that of the normal saline group at 14 - day (P 〈 O. 05), and the same situation showed up in the high - dose group at 35 - day. The activity of AKP in the high - dose group was lower than that of the normal saline group at 7 - day ( P 〈 0.05 ) . Conclusions In the experimental condition, apigenin could cause activity change of some testicular marker enzymes.
