摘    要:目的:探讨不同低氧浓度暴露对慢性疲劳综合征小鼠学习记忆能力及情绪的影响。方法:雄性ICR小鼠72只,随机分为6组,每组12只,即正常对照组(C0组)、慢性疲劳综合征建模组(CFS0组)、低氧干预正常对照组(C组)、慢性疲劳综合征低氧干预对照组(CFS组)、慢性疲劳综合征+15%氧浓度低氧暴露组(15%H组)和慢性疲劳综合征+18%氧浓度低氧暴露组(18%H组),其中CFS0组、CFS组、15%H组和18%H采用束缚和强迫游泳方式建立慢性疲劳综合征小鼠模型,C0组和C组同等条件下正常饲养。建模完成后,C0组和CFS0组立即处死,用于检验建模效果。C组继续饲养7天,CFS组自然恢复7天,15%H组和18%H组分别在15%和18%氧浓度环境暴露7天(1小时/天)。采用水迷宫、旷场测试、O迷宫测试及悬尾测试评定各组小鼠学习记忆能力及情绪变化。结果:(1)水迷宫结果显示,CFS0组潜伏期显著长于C0组(P﹤0.05),在平台所在象限的时间显著短于C0组(P﹤0.05)。低氧干预后,重复测量方差分析显示各组潜伏期明显不同;15%H组在平台所在象限时间与C组无显著性差异,但CFS组和18%H组在平台所在象限时间仍显著低于C组(P﹤0.05)。(2)CFS0组与C0组,C组、CFS组、15%H组及18%H组旷场、悬尾及O迷宫测试结果均无显著性差异(P﹥0.05);低氧干预后,各组行为学测试结果均无显著性差异(P﹥0.05)。结论:通过束缚和游泳方式可建立以空间学习记忆能力下降为特征的慢性疲劳综合征小鼠模型,在氧浓度为15%环境中暴露7天(1小时/天)明显改善慢性疲劳综合征小鼠学习记忆能力。

Abstract:Objective To study the effect of hypoxic exposure on spatial learning,memory capacity and emotions of mice with chronic fatigue syndrome(CFS).Methods 72 ICR male mice were randomly divide into 6 groups:normal group(C0),chronic fatigue syndrome group(CFS0),normal group with hypoxic exposure(C),chronic fatigue syndrome group without hypoxic exposure(CFS),chronic fatigue syndrome group with 15% O2 exposure(15% H) and chronic fatigue syndrome group with 18% O2 exposure(18%H).CFS was induced by forced swimming.Mice in groups 15%H and 18%H received hypoxic exposure one hour per day for seven days.Morris water maze(MWM),open-field test,zero maze test and tail suspension test(TST) were used to assess the emotion,spatial learning and memory capacity of mice.Results(1)MWM revealed that the escape latency and quadrantic time in group CFS 0 significantly decreased(P﹤0.05 and P﹤0.05),as compared to group C 0.There was significant difference in escape latency among different groups after hypoxic intervention.The time spent in target quadrant of groups CFS and 18% H was significantly shorter than that of group C(P ﹤ 0.05),while no difference between groups 15%H and C.(2)There were no significant differences in open-field test,zero maze test and TST between groups C 0 and CFS 0(P﹥0.05),as well as among the groups C,CFS,15%H and 18%H(P﹥0.05).Conclusion Hypoxic exposure 1 hour per day for seven days is helpful to improve the spatial learning and memory capacity in mice with CFS.
