趋化因子CXCL9在结直肠癌中的表达及其临床意义

摘    要:目的:分析趋化因子CXCL9[chemokine(C-X-C motif)ligand 9]在结直肠癌中的表达及其临床意义。方法:回顾分析复旦大学附属上海市第五人民医院手术治疗的60例结直肠癌患者的临床资料,采用免疫组织化学方法和实时荧光定量聚合酶链反应(Real-Time polymerase chain reaction,Real-Time PCR)技术检测结直肠癌组织、癌旁组织、区域淋巴结及正常结直肠组织中CXCL9的蛋白和mRNA的表达情况,采用酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测60例结直肠癌患者及20例健康志愿者的血清CXCL9浓度,探讨CXCL9与结直肠癌临床病理因素间的关系。结果:结直肠癌组织中CXCL9蛋白的阳性表达率及CXCL9 mRNA相对表达量均显著高于正常结直肠组织、癌旁组织、无癌转移的区域淋巴结;有癌转移的区域淋巴结中CXCL9蛋白的阳性表达率及CXCL9 mRNA相对表达量显著高于无癌转移的区域淋巴结,差异有统计学意义(P0.01)。CXCL9的表达与结直肠癌的浸润深度、淋巴结转移及Dukes分期相关(P0.05),而与年龄、性别、肿瘤直径无关(P0.05)。血清CXCL9浓度随Dukes分期的增加而升高(P0.05),有淋巴结转移的结直肠癌患者血清CXCL9浓度显著低于无淋巴结转移患者(P0.05)。结论:趋化因子CXCL9可能参与结直肠癌的发生、发展过程,其表达水平与结直肠癌的浸润深度、临床分期、转移等因素相关。

Abstract:Objective:To investigate the expression of CXCL9[chemokine (C-X-C)ligand 9] in patients with colorectal cancer (CRC).Methods:The clinical datum of 60 patients with CRC who received surgical treatment in the Fifth People's Hospital of Shanghai,Fudan University,were retrospectively analyzed.The expression of CXCL9 protein and mRNA in primary lesions of CRC,peritumoral tissues,regional lymph nodes and normal colorectal tissues were detected by immunohistochemistry and Real-Time polymerase chain reaction(PCR).Enzyme-linked immunosorbent assay(ELISA) was performed to detect the serum concentration of CXCL9 in 60 patients with CRC and 20 healthy volunteers.The relation between the expression of CXCL9 and clinic pathologic features of CRC was explored.Results:The positive rate of CXCL9 protein and relative expression of CXCL9 mRNA in primary CRC lesions were significantly higher than those in peritumoral tissues,regional lymph node without metastasis and normal colorectal tissues.The positive rate of CXCL9 protein and relative expression of CXCL9 mRNA in regional lymph node tissue with metastasis was significantly higher than that in regional lymph node tissue without metastasis,and the difference had statistical significance (P<0.01).The positive expression of CXCL9 was correlated to invasion depth,lymph node metastasis and Dukes stage(P<0.05),however,not correlated to age,gender and tumor diameter(P>0.05).The serum concentration of CXCL9 increased with the progression of Dukes stage(P<0.05),and the serum concentration of CXCL9 in patients with CRC with lymph node metastasis was significantly lower than that in patients without lymph node metastasis(P< 0.05).Conclusions:CXCL9 may play a role in the occurrence and progression of CRC,and the expression level of CXCL9 is correlated to invasion depth,clinical stage and metastasis of CRC.
