补肝散对感染血吸虫小鼠肝损伤后肝组织Bcl-2、Bax水平的影响

目的:观察补肝散干预感染血吸虫小鼠肝损伤模型后,对肝组织凋亡相关基因Bcl-2和Bax蛋白表达水平的影响,探讨补肝散对肝损伤的防护作用机制.方法:选择健康8周龄雄性昆明小白鼠108只,随机分为5组,将血吸虫尾蚴(18条/只)经小鼠腹部皮肤攻击感染造模,每组定期随机抽取6只小鼠,处死后留取其肝脏左叶,置于10％中性甲醛(用PBS配)固定,免疫组化法检测Bcl-2、Bax蛋白的表达水平.结果:①Bcl-2、Bax表达在模型组小鼠肝组织中的阳性率均明显升高,与正常组比较差异有显著性意义(P＜0.05),Bcl-2/Bax比值下降(P＜0.05)；②补肝散组与模型组相比小鼠Bel-2表达升高差异有显著性意义(P＜0.05),Bax表达升高差异无显著性意义(P＞0.05),Bcl-2/Bax比值增高差异有显著性意义(P＜0.05).结论:补肝散可促进Bcl-2表达,使Bcl-2/Bax比值上调,减少肝细胞的凋亡,进而防护血吸虫性小鼠肝损伤,发挥抗纤维化的作用.

Effects of Bugan powder on expression of Bcl-2 and Bax in mice with schistosome

Objective:From the aspects of pathological and apoptosis related genes Bcl-2 and Bax protein expression level,to research the intervention mechanism of Bugan powder on liver injury induced by schistosome in mice.Methods:One hundred and eight 8W old Kunming male mice were randomly divided into five groups,6 mice were selected and killed from each group periodically and randomly their liver specimen were kept placed in 10% neutral formalin to be fixed.The expression of Bcl-2 and Bax were detected by immunohistochemisty methods.Results:The expression of Bcl-2 and Bax in the model group were higer than those in the normal group(P<0.05).The expression of Bcl-2 in the Bugan group higher than those in the model group(P<0.05),the expression of Bax rised no significant differences(P>0.05),and Bcl-2/Bax increased significantly(P<0.05).Conclusion:Bugan powder can promote the expression of Bcl-2,so as to increase Bcl-2/Bax ratio,reduce liver cell apoptosis,intervene schistosomal liver injury,and play a role in anti-fibrosis.
