摘    要:目的探索亲和层析的方法,采用有效的方法提高几丁质树脂的使用效力和使用次数,降低成产成本使之更有利于提高产量。方法考察几丁质纯化的过程,探索出如何提高几丁质珠的重复使用次数,寻找出适合工业生产的纯化步骤。结果用高浓度的碱液进行洗柱,并延长浸泡时间到1h,可以使几丁质珠使用次数由原来的4~5次增加到7~8次。结论实验结果证实采用提高碱液浓度并延长浸泡时间可以增加几丁质树脂的使用次数,有利于指导实验室实验和药物开发。

Abstract:Objective Explore affinity chromatography method,the adoption of effective means to enhance chitin resin effectiveness and the use of frequency of use,lower-cost production to make it more conducive to raising output.Methods Investigation chitin purification process,and explore how to improve the chitin beads repetition frequency of use,finding suitable industrial production and purification steps.Results With a high concentration of caustic washing column,and the extension of immersion time to1h,can make use of chitin beads from 4-5 times the number increased to 7~8 times.Conclusions The experimental results confirmed using lye improve concentration and the extension of immersion time chitin can increase the frequency of use resin,to guide the laboratory experiments and drug development.
