胃癌中Runx3表达及其甲基化

目的探讨Runx3表达及其甲基化与胃癌的关系。方法采用逆转录-聚合酶链反应（RT-PCR）检测42例胃癌组织、癌旁正常组织及转移淋巴结中Runx3 mRNA的表达,同时用甲基化特异性PCR（MSP）方法检测Runx3甲基化状况。结果胃癌组织、转移淋巴结、癌旁正常组织中Runx3 mRNA的表达相对值分别为0.569±0.131、0.609±0.141、0.871±0.237,胃癌组织、转移淋巴结与癌旁正常组织相比,均有显著性差异（P〈0.05）。胃癌组织、转移淋巴结Runx3甲基化阳性率分别为71.4%、62.5%,两者无显著性差异（P〉0.05）;而癌旁正常组织中未检测到Runx3甲基化。胃癌组织中Runx3甲基化表达与肿瘤的分化程度有关,与性别、肿瘤大小、浸润深度等临床病理特征无关。结论Runx3甲基化是胃癌发生、发展过程中的频繁事件,通过对其检测,可能会对胃癌的早期诊断、治疗提供重要依据。

Expression of Runx3 Gene and Its Methylation in Gastric Carcinoma

Objective To investigate the expression and methylation status of Runx3 gene in gastric cancer. Methods The expression of Runx3 gene in 42 gastric carcinoma specimens,the adjacent nontumorous tissues and metastatic lymph nodes were detected by RT-PCR technique. Meanwhile,methylation-specific PCR was used to detect methylation status of Runx3 gene. Results The expression of Runx3 gene mRNA detected in gastric carcinoma 0. 569±0. 131 and metastatic lymph nodes 0. 609 ± 0. 141 were significantly lower than that in adjacent nontumorous tissues （0. 871±0. 237, P〈0. 05）. Aberrant methylation of Runx3 was detected in 71.4% of gastric cancer specimens, 62. 5% of metastatic lymph nodes, which showed no significant differences （P〉0.05）,and no methylation of Runx3 gene was found in the adjacent nontumorous tissues. Runx3 methylation was significantly correlated with the differentiation degree, but not with the gender or tumor size and invasion depth of gastric cancer. Conclusions Aberrant methylation of Runx3 gene is a common event in the occurrence and progression of gastric cancer, which may provide useful information for the early diagnosis and therapy.
