皮肤基底细胞癌组织中血管内皮细胞生长因子受体-2的表达

目的 研究血管内皮生长因子受体-2(VEGFR-2)在皮肤基底细胞癌(BCC)组织中的表达情况.方法 提取BCC组织的mRNA,以RT-PCR法检测标本中VEGFR-2mRNA的表达.同时应用蛋白质印迹法检测VEGFR-2蛋白的表达,免疫荧光法检测VEGFR-2在BCC组织中的定位.结果 BCC组织标本mRNA和蛋白水平中均可以检测到VEGFR-2的表达,并较正常人对照有显著增加.VEGFR-2在BCC细胞主要表现为膜性分布.结论 皮肤BCC组织细胞能够高异常表达VEGFR-2,可能是导致BCC细胞肿瘤性增殖的原因之一.

Expression of vascular endothelial growth factor receptor-2 in the tissue of basal cell carcinoma

Objective To determine the expression of vascular endothelial growth factor receptor-2 (VEGFR-2) in the tissue of basal cell carcinoma(BCC).Methods Tissue specimens were collected from 7 patients with BCC and healthy human controls,mRNA was extracted from the specimens.RT-PCR assay was employed to determine the mRNA expression of VEGFR-2,immunofluorescence and Western blotting methods to detect the distribution and protein expression of VEGFR-2 respectively in the tissue specimens.Results Both the mRNA and protein expressions of VEGFR-2 were observed and significantly higher in the tissue specimens of BCC than those in the normal controls(1.171±0.165 vs 0.489±0.143, 0.341±0.022 vs 0.258±0.020).The VEGFR-2 protein was mainly located in the membrane of BCC cells. Conclusions The expression of VEGFR-2 is abnormally high in BCC tissues,which may contribute to the proliferation of BCC cells.
