实验性大鼠颅脑损伤后GFAP的表达研究

摘    要:目的:通过建立针刺颅脑损伤动物模型,研究颅脑损伤后星形胶质细胞随损伤时间的变化规律.方法:采用针刺法复制大鼠脑挫伤模型,造成右顶叶局限性脑挫伤,于伤后不同时间将其处死,取脑组织,用免疫组化方法及图像分析技术检测脑组织中GFAP的含量变化,并与正常对照组进行比较.结果:GFAP随损伤时间会出现规律性变化,呈现上升后下降的趋势,伤后3天表达达最高,伤后21天时基本恢复正常.且各时间段双侧大脑表达无明显差异.结论:在针刺法致脑损伤模型中星形胶质细胞参与了颅脑损伤后的修复过程,其标志性蛋白GFAP随损伤时间会出现规律性改变.

Abstract:Objective: To study the expression of glial fibriliary acidic protein(GFAP) after brain injury and establish brain injury animal model through stab wound. Methods: The preliminary study were performed on the experimental rat's brain tissue 0.5h-21d after the injury by using the immunohistochemical method. The results were compared with the normal animal. Results: The results revealed that the expression of GFAP was increased at first reached a peak at 3 day, then decreased, at 21 day it was back the normal level. Conclusion: It suggested that regular expression of GFAP would appear after brain injury, astrocyte had taken part in the repair of stab wound brain injury.
