Correlations of the expressions of BMI-1 gene and human epidermal growth factor receptor-2 in breast cancer with micrometastasis in peripheral blood

Abstract:Objective To observe the positive expressions of BMI-1 and human epidermal growth factor receptor-2(HER-2)in breast cancer tissues of patients with breast cancer,and to investigate the correlations of BMI-1 and HER-2 in breast cancer with micrometastasis in peripheral blood.Methods Real-time fluorescence quantitative PCR was used to detect the relative expression of human mammaglobin mRNA in 89 patients with breast cancer,by which 89 patients were divided into 41 patients with micrometastases in peripheral blood(micrometastasis group)and 48 patients with no micrometastasis(non-micrometastasis group).The serum levels of hypoxia inducible factor-1 alpha(HIF-1α)and matrix metalloproteinase-9(MMP-9)as well as the positive rates of BMI-1 and HER-2 in breast cancer tissues were compared between two groups.Multivariate logistic regression analysis was used to analyze the influencing factors of micrometastasis in peripheral blood of patients with breast cancer.Results The serum levels of HIF-1αand MMP-9 as well as the positive rates of BMI-1 and HER-2 were higher in micrometastasis group(52.32(49.61,59.12)ng/L,50.12(46.95,55.74)μg/L,78.05%,85.37%)than those in non-micrometastasis group(47.89(43.12,53.41)ng/L,48.12(43.21,51.06)μg/L,43.75%,47.92%)(P<0.05).The serum HIF-1α(OR=1.780,95%CI:1.695-1.876,P=0.003),MMP-9(OR=1.611,95%CI:1.521-1.913,P=0.004),BMI-1(OR=7.000,95%CI:2.314-21.179,P=0.001)and HER-2(OR=9.666,95%CI:3.225-28.971,P<0.001)were the influencing factors of micrometastasis in peripheral blood of patients with breast cancer.Conclusion The levels of BMI-1 and HER-2 increase in patients with breast cancer,and are associated with micrometastasis in peripheral blood of patients with breast cancer.
