MMP2在非小细胞肺癌中的表达及其与肿瘤微血管密度的关系

摘    要:目的探讨MMP2在非小细胞肺癌组织中的表达及其与肿瘤微血管密度(MVD)的关系。方法用链菌素生物素蛋白一过氧化物酶免疫组织化学法(SP法),检测83例手术切除的非小细胞肺癌组织及其它们的交界区组织和24例正常组织中MMP2的表达,分析MMP2在肿瘤组织中的过表达与非小细胞肺癌的临床病理特征的关系。用CD34标记肿瘤微血管,分析MMP2的过表达与肿瘤MVD的关系。结果MMP2主要表达于肿瘤细胞的胞浆,在癌旁交界区组织也有表达,肿瘤组织的过表达率显著高于交界区(P〈0.005),在正常组织中不存在过表达;MMP2的过表达与肿瘤类型、病理分级、性别、年龄、吸烟史无明显关系;MMP2在肿瘤组织的过表达率与淋巴结转移状态及临床分期有关(P〈O.05)。肿瘤组织中MMP2过表达组的MVD显著高于非过表达组。结论MMP2参与了非小细胞肺癌发生发展过程,并且与肿瘤血管生长有关系,MMP2与淋巴结转移和临床分期密切相关。

Abstract:Objective To investigate the relationship between MMP2 expression and microvesseldensity in non-small-cell lung carcinoma.Methods MMP2 expressions were determined in surgically treated 83 cases of non-small cell lung carcinoma tissues and their adjacent tissues and 24 cases of normal lung tissues by streptavidinperoxdase immunohistochemical technique.The microvessel of tumor tissue were indicated by CD34 antibody.Results The immunoreactivities of MMP2 were mainly shown in cytoplasma of tumor cells.The overexpressions of MMP2 in carcinoma tissues were significantly higher than that in adjacent tissues(P<0.005).No overexpression was observed in normal tissues.Their overexpressions were not related to tumor types,clinical stages,pathological grades,sex,age,smoking history.The overexpression of MMP2 in cancer tissues was related to clinical stages and lymph node metastasis status(P<0.05).The number of microvessel in tumor tissue with the overexpresion of MMP2 was more than that without overexpresion.Conclusion MMP2 may take part in the progress of non-small-cell lung carcinoma and the growth of tumor tissue microvessel.The overexpression of MMP2 in cancer tissues is related to clinical stages and lymph node metastasis status.
