食管鳞癌组织中Nrf2和Bcl-2蛋白表达的关系及其临床意义

摘    要:目的: 探讨核因子E2相关因子2(Nrf2)及Bcl-2蛋白在食管鳞癌组织中的表达及其与临床病理指标之间的关系。方法:采用免疫组织化学方法(SP法)和Western blot技术分别检测85例食管鳞癌组织及距食管癌组织边缘5 cm以上的35例正常食管组织中Nrf2及Bcl-2蛋白的表达,并分析两者之间的相关性及其与食管癌临床病理指标之间的关系。结果:免疫组织化学结果显示,Nrf2和Bcl-2蛋白在食管鳞癌组织的表达阳性率(分别为83.5%和72.9%)均较正常食管组织(分别为14.2%和5.7%)明显升高(P<0.01);Western blot结果显示,Nrf2和Bcl-2蛋白在食管鳞癌组织相对表达量(分别为0.855±0.078和0.801±0.075)均较正常食管组织(分别为0.135±0.011和0.173±0.040)明显升高(P<0.01)。Nrf2和Bcl-2蛋白表达均与食管鳞癌组织中的病变长度、浸润深度、分化程度及淋巴结转移密切相关(P<0.05),但与食管鳞癌患者病变部位及病变类型无显著相关(P>0.05)。Spearman相关分析显示:食管鳞癌组织中Nrf2与Bcl-2蛋白表达呈显著正相关(r=0.515,P<0.01)。结论:Nrf2和Bcl-2的表达可以作为判断食管鳞癌恶性潜能的重要生物学指标。

Abstract:OBJECTIVE:To investigate the expressions of Nrf2 and Bcl-2 and clinical pathological features in esophageal carcinoma tissues. METHODS:Immunohistochemistry (SP) and Western blot were used to detect the expressions of Nrf2 and Bcl-2 in 85 specimens of esophageal carcinoma tissues and normal esophageal tissues (5 cm distant to the margin of esophageal cancer). RESULTS:The positive rates of Nrf2 and Bcl-2 protein(83.5%and 72.9%, respectively) in esophageal carcinoma tissues were both higher than normal tissues(14.2% and 5.7%, respectively)(P〈0.01). The relative contents of Nrf2 and Bcl-2 protein(0.855±0.078 and 0.801±0.075,respectively) in esophageal carcinoma tissues were both higher than normal tissues (0.135±0.011 and 0.173±0.040,respectively) (P〈0.01). There was a significant difference in Nrf2 protein expressions between lesion length,depth of invasion,histological grade and lymph node metastasis (P〈0.05),but not correlated with lesion location and general classification of tumors (P〉0.05). Spearman analysis showed that Nrf2 protein expression was positively correlated with Bcl-2 protein expression (r=0.515,P=0.000). CONCLUSION:Nrf2 and Bcl-2 protein expression could be considered as biological indicators of the malignant potential of esophageal squamous carcinoma.
