LncRNA-UCA1与淮安食管癌发病风险的初步研究

摘    要:目的: 初步筛选淮安食管鳞癌与癌旁组织中差异表达的LncRNAs,为进一步研究lncRNAs在食管鳞癌发病机制中的作用奠定基础。方法:分别提取3例患者食管癌组织及配对癌旁组织的总RNA,体外逆转录制备并标记为双链cDNA(ds-cDNA),与人类LncRNA表达谱芯片进行杂交,经扫描后,用Agilent GeneSpring GX v11.5.1软件进行分析运算,筛选出差异表达的LncRNAs。qRT-PCR分析LncRNA在食管癌细胞及组织中的表达情况。结果:按照倍数≥2.0、P≤0.05的标准,筛选出表达差异有统计学意义的LncRNAs共1 019个,其中399个上调,620个下调。GO分析和通路分析结果显示这些LncRNAs调控的差异表达的mRNAs主要与细胞周期、细胞分化、蛋白酶的活动等有关。结合Gibb食管癌相关的LncRNA差异表达谱分析发现lncRNA-UCA1可能参与食管癌发生发展。qRT-PCR结果显示,与永生化食管上皮细胞及癌旁组织相比,食管癌细胞及食管癌组织中lncRNA-UCA1均表达下调。结论:共筛选出淮安食管鳞癌与癌旁组织中1 019个差异表达LncRNAs,其中LncRNAs-UCA1可能成为食管癌早期发现和诊断的生物标志物。

Abstract:OBJECTIVE:To analyze LncRNA differential expression in tumoral and normal-paired tissues of esophageal squamous cell carcinoma in Huaian and to explore the role of tumor-associated LncRNAs in the pathogenesis of esophageal squamous cell carcinoma. METHODS:Total RNA was extracted from 3 patients' esophageal cancer tissues and corresponding adjacent normal tissues. Then,each sample was amplified and transcribed into fluorescent cRNA with a random priming method. The labeled cDNAs were hybridized onto the Human LncRNA Array v2.0 (8×60K,Arraystar). After being scanned,Agilent GeneSpring GX v11.5.1 was used to screen the differentially expressed LncRNAs. qRT-PCR was used to analyze LncRNA expression in cells and tissues of esophageal cancer. RESULTS:Among the 1 019 significantly differentially expressed LncRNAs (Fold Change≥2.0,P≤0.05),there were 399 LncRNAs overexpressed and 620 under-expressed. GO analysis and Pathway analysis showed that the differentially expressed mRNAs,regulated by LncRNAs,were mainly associated with the cell cycle,cell differentiation, protease activities etc. Compared to LncRNA differences related to the Gibb's esophageal expression profile analysis,LncRNA-UCA1 might participate in esophageal cancer development. qRT-PCR results showed that LncRNA-UCA1 was significantly under-expressed in esophageal cancer cells and esophageal cancer tissues when compared with immortalized esophageal epithelial cell and adjacent normal tissues. CONCLUSION:A total of 1 019 significantly differentially expressed LncRNAs were identified,and LncRNA-UCA1 may be a potential biomarker in the early detection and diagnosis of esophageal cancer.
