酸敏脂质体的制备及其在肠缺血再灌注小鼠体内重要脏器中的分布研究

摘    要:目的:比较4种制备脂质体的方法,观察酸敏脂质体包裹的碘125-白介素-8(^125I-IL-8)在肠缺血-再灌注损伤小鼠体内的分布情况。方法:用4种不同的的方法制备包裹^125I-IL-8的脂质体,比较其包封率。在肠缺血-再灌注损伤小鼠尾静脉注射包裹^125I-IL-8的酸敏脂质体,通过检测^125I的计数,观察酸敏脂质体在各重要脏器中的分布情况。结果:反相蒸发法与钙融合法联合制备的酸敏脂质体有较高的包封率。酸敏脂质体与普通脂质体相比,在受损伤脏器部位有较高分布。结论:酸敏脂质体在损伤部位(pH较低处)有相对较高的分布。

Abstract:Objective:To compare the four kinds of methods of making liposomes,and to observe the tissue distribution of pHsensitive liposome encapsulated 125 IIL8 in intestine after ischemiareperfusion in mice.Methods:Encapsulated 125 IIL8 was made with four kinds of liposome and their envelope ratios were compared.After an ischemiareperfusion episode of intestine,pHsensitive liposome encapulated 125 IIL8 was injected into tail of mice,and the tissue distribution of pHsensitive liposomes was observed.Results:The liposomes made by calciuminduced fusion and reversephase evaporation vesides had the highest envelope ratio.The distribution of pHsensitive liposomes was more abundant than that of ordinary liposomes in the injured tissue.Conclusions:The denser distribution of pHsensitive liposomes could be observed in injured tissue with a lower pH.
