薯蓣皂苷元对移植人低分化胃腺癌荷瘤裸鼠肿瘤重量的影响

摘    要:目的 :通过动物整体实验 ,观察薯蓣皂苷元 ( diosgenin,Dio)对移植人低分化胃腺癌细胞株 ( MGC- 80 3)荷瘤裸鼠肿瘤增重的影响 ,为确定该药物的临床抗肿瘤适应证寻求依据。方法 :将左侧腋部皮下已植入 1 mm× 1 mm× 1 mm左右 (重量约 40 mg) MGC- 80 3肿块的裸鼠随机分为对照组、环磷酰胺组及 3个不同剂量 Dio组。给药周期为 6周。给药结束 ,将动物脱臼处死 ,剥离出肿块称重 ,比较各组肿瘤重量 ,并求出肿瘤抑制百分率。结果 :Dio 40 0、2 0 0和 1 0 0 mg· kg-1ig对移植MGC- 80 3荷瘤裸鼠肿瘤生长均具有明显的抑制作用。并呈现出较好的量效关系 ,平均抑瘤率可达44.9%~ 64.5 %。结论 :Dio对 MGC- 80 3增长具有抑制作用

Abstract:Objective: To examine the effect of diosgenin (Dio) on tumor growth of nude mice bearing human gastric cancer (MGC 803) xenograft in vivo, and to find the evidence of clinical indication of Dio.Methods: Human gastric cancer (MGC 803) xenograft in size 1 mm×1 mm×1 mm (about 40 mg) were transplanted into left axillary hypoderm of nude mice. In 6 week period, Dio was given via oral perfusion in dose of 400,200 and 100 mg·kg -1 every day;Cytoxan was given via oral perfusion in dose of 25 mg·kg -1 ,every other day; animals in control group were given distilled water via oral perfusion in volume of 20 ml·kg -1 every day. Results: Dio ig in dose of 400,200 and 100 mg·kg -1 possess significant suppressive effect on tumor growth, and showed better dose dependent relationship. Average inhibition rates were 44.9%~64.5% .Conclusion: Dio possess significant suppressive effect on human gastric cancer(MGC 803) xenograft.
