Clusterin、bcl-2及p53在前列腺上皮组织中的表达及相关性研究

摘    要:目的通过检测新的凋亡抑制因子Clusterin在前列腺癌组织中的表达,探讨其与前列腺癌发生发展的关系及其与bcl-2和p53表达的关系.方法采用免疫组织化学染色法检测Clusterin、bcl-2、p53在10例正常前列腺组织、15例良性前列腺增生组织(BPH)、49例前列腺癌组织标本中的表达.结果Clusterin在前列腺正常、增生、癌组织中的阳性及弱阳性表达率分别为10%(1/10)、66.6%(10/15)、91.8%(45/49).前列腺癌组织中Clusterin表达水平明显高于前列腺正常组织(P<0.005)及增生组织(P<0.005),且在癌组织中与肿瘤临床分期(P<0.001)、病理分级(P<0.001)呈正相关.前列腺癌Clusterin表达与bcl-2(P<0.05)及p53(P<0.001)表达呈正相关关系.结论Clusterin基因可能通过抑制凋亡在前列腺癌的发生发展中发挥重要作用,且其表达与bcl-2、p53的异常表达密切相关.

Abstract:Objective To investigate the expression of an inhibitor gene of apoptosis, clusterin, in prostate cancer and its relationship with the genesis and progression of prostate cancer and with the expressions of bcl 2 and p53. Methods The expressions of clusterin, bcl 2, and p53 in 10 cases of normal prostatic tissues, 15 cases of benign prostatic hyperplasia (BPH), and 49 cases of prostate cancer were examined by immunohistochemical staining. Results The rates of strong positive or weak positive of clusterin in normal prostatic tissues, BPH, and prostate cancer were 10% (1/10), 66.6% (10/15), and 91.8% (45/49), respectively. The expression level of clusterin in prostate cancer tissues was significantly higher than that in normal prostatic tissues ( P <0 005) and BPH ( P <0 005), and was positively correlated with clinical stages (P<0.001) and pathological grades ( P <0 001) of prostate cancer. The expression level of clusterin in prostate cancer was positively correlated with bcl 2 expression ( P <0 05) and p53 expression ( P <0 001). Conclusion Clusterin, closely associated with bcl 2 and p53, may play an important role in the genesis and progression of prostate cancer through the inhibition of apoptosis.
