地塞米松诱导体外培养星型胶质细胞凋亡机制的研究

摘    要:目的:探讨地塞米松引起体外纯化培养的大鼠大脑皮质星型胶质细胞凋亡的机制。方法:不同浓度的地塞米松(浓度为10-5、10-4、10-3m o l/L)与纯化培养的大鼠大脑皮质星形胶质细胞共同孵育24小时后,用免疫细胞化学方法检测P 53、Bax及B cl-2在星型胶质细胞内的表达情况,表达的强弱用平均光密度表示。结果:P 53和Bax的表达随着地塞米松浓度的升高而增强,与对照组比较,各组均有极显著性差异(P<0.01),B cl-2的表达随着地塞米松浓度的升高先是增强而后减弱,与其他组比较10-3组有显著性差异(P<0.05)。结论:P 53和Bax的表达增强可能是大剂量地塞米松引起星型胶质细胞凋亡的主要因素。

Abstract:Objective: To study the mechanism of apoptosis of astrocytes induced by Dexamethasone.Method: Pure cultured astrocytes from newborn rat cerebral cortex were incubated with different concentrations of Dexamethasone for 24 hours,then the expression of P53,Bax and Bcl-2 in astrocytes was measured by immunocytochemical analysis.Results: The immunoreactivity for P53 was markedly increased with the elevation of concentration of Dexamethasone;the ratio of Bax expression to Bcl-2 in 10~(-3) group was significantly increased compared with other groups.Conclusion: The enhanced expression of P53 and Bax may contribute to the apoptosis of astrocytes in vitro induced by vast amount of Dexamethasone.
