酸味中药复方对2型糖尿病大鼠细胞凋亡及相关基因表达的影响

目的观察酸味中药复方对2型糖尿病大鼠不同组织细胞凋亡的影响。方法高热量饲料加小剂量链脲佐菌素(STZ)腹腔注射建立2型糖尿病大鼠模型,分别用酸味、甘味、苦味中药复方和二甲双胍干预,在给药12周后,检测大鼠胰腺、心脏、肾脏凋亡细胞及其相关基因Bax、Bcl-2的蛋白表达。结果酸味中药复方可抑制胰腺、心脏、肾脏的细胞凋亡,上调Bcl-2/Bax比值,与模型组比较有显著性差异(P<0.05)。结论酸味中药复方对2型糖尿病模型大鼠靶器官的保护作用,可能与调节Bax、Bcl-2的蛋白表达,从而降低靶器官的细胞凋亡有关。

Effect of Sour-flavor Chinese Herbal Formula on Cell Apoptosis and Gene Expression in Diabetes Mellitus Type 2 Rat Model

Objective To observe the effect of sour- flavor Chinese compound on cell apoptosis in diabetes mellitus type 2 (T2DM) rat model. Methods T2DM rat model was established by feeding with high- calorie forage and intraperitoneal injection of streptozotocin. The model rats were randomly divided into 5 groups: the model group and four treatment groups which were given sour- flavor Chinese compound, sweet- flavor Chinese compound, bitter- flavor Chinese compound and dimethyldiguanide respectively. Meanwhile, 6 normal rats were used as the blank control. After 12 weeks treatment, cell apoptosis in pancreas, heart and kidney was evaluated and the protein expression of Bcl- 2 and Bax was detected by immunohistochemistry method. Results Sour- flavor Chinese compound could inhibit the cell apoptosis in pancreas, heart and kidney and increase significantly the Bcl- 2/Bax ratio( P < 0.05 as compared with the model group) .Conclusion The protective effect of the sour- flavor Chinese compound on the target organs of T2DM rat model is probably related to the regulation of the expression of Bcl- 2 and Bax gene and the decrease of cell apoptosis in target organs .
