摘    要:目的:研究p53基因和顺铂联合应用对胆管癌细胞的作用。方法:将重组体腺病毒p53和顺铂联合作用于人胆管癌细胞QBC939,对p53基因的表达、细胞的生长抑制及机制进行分析。结果:用Ad-LacZ进行复组腺病毒转导效率的检测,发现当MOI为100以上时,重组腺病毒可使90%以上的培养的人胆管癌QBC939细胞被传导,用RT-PCR方法,在胆管癌QBC39细胞系中p53无表达。重组体腺病毒能介导外源基

Abstract:Objective To study the effect of combination ap plication p53 gene an d cisplatine in QBC939 bile duct cancer cell lines. Methods Ana lyse the express ion of p53 gene and inhibited growth of QBC939 cell lines by combination applica tion p53 gene mediated by recombination adenoviral vector and cisplatine. Result s If MOI>100, 90% QBC939 cells were transductd by recombination adenovi ral vect or when we detected the efficiency of transduction by using Ad-LacZ system. The expression of p53 gene was not detected by RT-PCR method. The high active expres sion of p53 gene in bile duct cancer cell lines used recombination adenoviral ve ctor. The expression of p53 gene by this way can inhibit growing and forming col ony of QBC939 cell lines. Combination application recombination adenoviral p53 g ene vector and cisplative can obviously inhibit QBC939 cell growth. By FCM analy sing, p53 gene can induce cells apoptosis and make it G1 arrest; Cisplatine can induce cell apoptosis and make it G2 arrest. Conclusions p53 ge ne can enhance the sensibility of cisplatine in QBC939 cell lines.
