麦冬有效部位对支原体感染小鼠肺组织黏蛋白MUC5ac表达的影响

摘    要:目的:通过实验研究麦冬有效部位对肺炎支原体(MP)感染小鼠的肺组织黏蛋白5ac(MUC5ac)表达及分泌的影响。方法:将100只BALB/c小鼠,分为正常对照组,模型组,阿奇霉素组,养阴清肺组,麦冬多糖高、中、低剂量组,麦冬皂苷高、中、低剂量组,每组10只。除正常组外,其余各组采用MP滴鼻法造模,连续造模3 d后,各组开始灌胃给药,每天1次,阿奇霉素组连续给药5 d,其他组连续给药6 d后,收集肺泡灌洗液(BALF),ELISA检测BALF中MUC5ac的蛋白含量;采集小鼠肺组织,PCR法测定肺组织中MUC5ac mRNA的表达。结果:1)肺组织MUC5ac mRNA的表达:与正常组相比,模型组肺组织MUC5ac mRNA的表达极显著上升(P<0.01);与模型组对比,阿奇霉素的表达量降低,具有极显著性(P<0.01);麦冬多糖高、中、低剂量组,养阴清肺组及麦冬皂苷高剂量组的表达量降低,具有显著性差异(P<0.05)。2)BALF MUC5ac的含量:与正常组对比模型组含量显著增加(P<0.01),与模型组对比,阿奇霉素组、麦冬皂苷高剂量组含量显著减少(P<0.01),麦冬皂苷中剂量组和麦冬多糖高、中剂量组含量减少,具有显著性差异(P<0.05)。结论:麦冬有效部位可下调MUC5ac蛋白的表达及分泌。

Abstract:Objective:To study the effect of Radix Ophiopogonis on the expression and secretion of MUC5ac in lung tissue of mice infected with Mycoplasma pneumoniae(MP).Methods:100 BALB/c mice were divided into normal control group,model group,Azithromycin group,Yangyin Qingfei group,Ophiopogon japonicus polysaccharide high,medium and low dose groups,Ophiopogon japonicus saponin high,medium and low dose groups,with 10 mice in each group.Except the normal group,the other groups were nasal dropped with MP for 3 days,once a day,then each group was given the corresponding medicine or saline continuously,Azithromycin for 5 days,the other groups for 6 days.The alveolar lavage fluid(BALF)was collected and the protein content of MUC5ac in BALF was detected by ELISA.The expression of MUC5ac mRNA in lung tissue was detected by PCR.Results:1)The expression of MUC5ac mRNA in lung tissue:Compared with the normal group,the expression of MUC5ac mRNA in the lung tissue of the model group was significantly increased(P<0.01),compared with the model group,the expression of Azithromycin was significantly decreased(P<0.01),the expression of MUC5ac mRNA in the high,medium and low-dose groups of Ophiopogon japonicus polysaccharide,Yangyin Qingfei group and Ophiopogonin saponin high-dose group was decreased with significant difference(P<0.05).2)MUC5ac in BALF:Compared with the model group,the content of MUC5ac in BALF of mice was significantly increased(P<0.01),compared with the model group,the content of MUC5ac in the Azithromycin group and Ophiopogon saponin high-dose group was significantly decreased(P<0.01),and the content of Ophiopogon saponin medium dose group,Ophiopogon japonicus polysaccharide high-dose group and middle dose group was significantly decreased(P<0.05).Conclusion:The effective fraction of Ophiopogon japonicus could down regulate the expression and secretion of MUC5ac protein.
