神经生长因子在哮喘小鼠骨髓嗜碱粒细胞的表达研究

摘    要:目的:检测正常和过敏性哮喘小鼠骨髓嗜碱粒细胞合成神经生长因子(NGF)的能力,探讨过敏性哮喘可能的发病机制。方法:建立过敏性哮喘小鼠模型,取哮喘小鼠及正常对照小鼠的骨髓嗜碱粒细胞,利用PCR技术检测嗜碱粒细胞NGF mRNA的表达量。结果:常规PCR示正常组和哮喘组小鼠骨髓嗜碱粒细胞均表达NGF;实时定量PCR示哮喘组小鼠嗜碱粒细胞NGF的表达高于正常组。结论:嗜碱粒细胞表达NGF,且在mRNA水平哮喘组小鼠嗜碱粒细胞NGF的表达高于正常组,提示嗜碱粒细胞是哮喘时NGF的重要来源。

Abstract:Objective: To investigated whether mouse basophils could express nerve growth factor(NGF) and to explore the pathogenesis of allergic asthma.Methods: The NGF mRNA of basophils from normal and allergic asthma mice bone marrow cells was measured by PCR.Results: PCR analysis showed that basophils from both normal and asthma mice expressed NGF,and the mRNA expression level of NGF in basophils from asthma mice was higher than that from normal mice.Conclusion: Basophils may be an important source of NGF in allergic asthma.
