心肌营养素-1在扩张型心肌病大鼠心肌的表达

目的探讨心肌营养素-1（CT-1）在DCM大鼠心肌的表达及作用。方法30只成年雄性Wistar大鼠随机分为两组：DCM组20只，腹腔内注射阿霉素（ADM）2．5mg／kg，2次／周，共6周，诱导大鼠DCM模型；对照组10只，同法注射等量生理盐水。第8周时行心脏超声检测，用VG染色观察心肌组织病理改变，用半定量RT—PCR检测CT-1在对照组大鼠与DCM大鼠心肌的表达。结果DCM组的心脏较对照组明显扩大，且心肌纤维化明显，心肌CT-1表达明显升高（P〈0．01）。结论在阿霉素诱导的DCM大鼠心肌中CT-1过度表达，可能与其保护心肌细胞、促进心肌细胞肥大有关。

Expression of Cardiotrophin-1 in the Dilated Cardiomyopathy of Rats

Objective We aim to investigate the effect and expression of cardiotrophin-1 （ CT-1 ） in the ventricle of rats with dilated cardiomyopathy. Methods Thirty adult male Wistar rats were randomly divided into the control group and the model group. The model group （ n = 20） was administered adriamyein intraperitoneally at a dose of 2.5mg/kg,twice a week for 6 weeks ,and the control group （ n = 10） was received an equivalent volume of 0. 9% saline alone intraperitoneally. Eehocardiographie measurements were obtained after 8 weeks. The pathological change was analyzed by histological Van Gieson stain. Later the expressions of CT-1 mRNA were tested with RT-PCR method. Results The hearts of the DCM group showed that the chamber of left ventricle was enlarged and myocardial fibrosis was more obvious than that of the control group;the expression of CT-1 mRNA increased obviously （P 〈0.01 ） as well. Conclusion The CT-1 mRNA was over-expression of rats with dilated cardiomyopathy in the adriamycin-induced, which may be relevant to protect myocardial cell and advance myocardial cell hypertrophy.
