附子和四逆汤表观药动学参数的测定

摘    要:以毒性为指标测定附子及四逆汤的表观药动学参数。方法为:先测小鼠ipLD_(50),另取小鼠ip首次剂量后分为不同间隔时间组,分别通过LD_(50)补量法测定间隔不同时间后首次注入药物的体存量,以log体存量对用药后间隔时间作图得药物体存量的经时性变化曲线,并计算药动学参数。经判断附子和四逆汤在体内的经时性变化均符合二室模型。计算结果二药分布相的半衰期分别为1.15h及0.35h,消除相的半衰期分别为17.0h及5.8h。

Abstract:Apparent pharmacokinetic parameters of Radix Aconiti Lateralis preparata and Si Ni Tang were determined by acute mortality of mice. l.The ip LD50 of mice was determined. 2.Mice received a dosage . (about LD10) were divided into 5-6 interval groups. For each group,ip LD50(t) was determined after appropriate interval.3.Residual of the first dosage after the interval of t =LD50-D50(t) . 4.on the basis of interval time and residual (log) relationship, a curve of dynamic change after initial dosage was drawn, and apparent pharmacokinetic parameters were calculated. The T1/2a of Radix Aconiti Lateralis preparata and Si Ni Tang were 1.15 h and 0.35 h, Ti/2B were 17.0 h and 5.8h respectively.
