葛根总黄酮对不同急性髓系白血病细胞株增殖抑制的影响

摘    要:为了探讨葛根总黄酮对急性髓系白血病(acute myeloid leukemia,AML)的作用及其可能的机制,观察不同浓度葛根总黄酮体外对4种AML细胞株Kasumi-1、HL-60、NB4和U937的增殖抑制影响和细胞周期的变化。应用MTT法检测细胞株细胞的增殖抑制作用,用流式细胞术检测细胞周期的变化。结果表明:一定浓度葛根总黄酮对4种AML细胞株增殖抑制作用差异显著,抑制强度依次为NB4细胞Kasumi-1细胞U937细胞HL-60细胞;葛根总黄酮不同程度地改变NB4、Kasumi-1、U937及HL-60细胞周期进程,使G1期细胞比例下降,S期细胞比例增高,出现亚二倍体峰,呈现浓度依赖性。结论:葛根总黄酮体外对不同AML细胞株具有选择性增殖抑制作用,以对NB4细胞株的影响最为显著。
