缺氧诱导分化因子-1对小鼠主动脉平滑肌细胞增生作用的研究

摘    要:目的探讨缺氧诱导分化因子-1对血管平滑肌细胞的致增生效应。方法采用改良贴块法培养小鼠主动脉平滑肌细胞,通过测定细胞甲基-3H胸腺嘧啶核苷(3H-TdR)掺入、MTT廓清和增殖细胞核抗原(PCNA)表达来评价不同浓度缺氧诱导分化因子-1对血管平滑肌细胞增生的影响。结果缺氧诱导分化因子-1呈剂量依赖性地增加平滑肌细胞的3H-TdR掺入、MTT廓清和PCNA的表达。结论缺氧诱导分化因子-1可促进血管平滑肌细胞增生。

Abstract:Objective To study the proliferative effects of FIZZ1(found in inflammatory zone 1) on cultured mouse vascular smooth muscle cells(VSMCs).Methods Methyl-3H thymidine(3H-TdR) incorporation,MTT cleaning and the expression of PCNA were determined on VSMCs after stimulation by FIZZ1.Results FIZZ1 caused marked increase of 3H-TdR incorporation,MTT cleaning and the expression of PCNA in VSMCs dose-dependently.Conclusion FIZZ1 dose-dependently stimulated the proliferation of VSMCs.
