人脐带间充质干细胞分化为神经细胞的形态学改变

摘    要:目的研究人脐带华尔通胶来源的间充质干细胞(MSCs)分化为神经细胞的形态学特征,为神经细胞移植探索细胞来源。方法用复方丹参注射液诱导人脐带来源的MSCs向神经细胞分化,用光学和电子显微镜对分化和未分化的细胞进行观察;了解细胞分化同形态学改变的关系。结果丹参可诱导人脐带MSCs向神经样细胞分化,光镜下细胞变短,体积变小,细胞形成双极或多极的细胞体,细胞呈星型,部分细胞的胞质收缩形成细胞的突起;扫描电镜下观察,复方丹参注射液诱导后,细胞胞体形成多样的神经样轴突,细胞之间形成网络样的连接;透射电镜下,丹参诱导后,细胞内结构、细胞器发生明显变化,细胞成熟,胞质内可见大量的线粒体,粗面内质网和游离的核糖体等,诱导后的细胞可见尼氏体样结构——大量排列的粗面内质网和广泛分布的游离核糖体和发达的线粒体;诱导后的细胞也表达神经细胞的标记。结论复方丹参注射液不仅诱导人脐带来源的MSCs发生神经样细胞的形态学改变,同时伴发有细胞分化的过程和神经细胞杯记表达,证明人脐带来源的MSCs具有分化为神经细胞的能力。

Abstract:Objective To study the morphologic characteristic of neuron-like cells of human umbilical cord-derived mesenchymal stem cells (MSCs). This study has been designed to may expand the potential role of MSCs in neurotransplantation and construction of tissue bank. Materials and Methods Composite salvia injection (CSI ) was used to induce the human umbilical cord-derived MSCs to differentiate respectively. The morphologic changes of differatiated and undifferentiated cells were compared with phase contrast microscopy, electron microscopy (EM) and transmission electron microscope (TEM). Results CSI can induce MSCs to differentiate to neuron-like cells. Found by phase contrast microscopy and electron microscopy (EM), the inducing treatments caused Wharton's jelly cells to undergo profound changes in morphology. The induced MSCs developed rounded cell bodies with multiple neurite-like extensions, finally, resembled bipolar or multipolar neurons, with processes that formed networks reminiscent of primary cultures of neurons. Observed by transmission electron microscope (TEM), their ultramicrostructure were more mature, with increasing cell organs and tigroid body similar to the neuron. Treatment with CSI can also induced MSCs to express neuronal cell markers. Conclusion Human umbilical cord derived MSCs were capable of differentiating into neuron-like cells with the treatment of CSI. The induced MSCs not only underwent nerve-cell like morphologic changes, ultramicrostructure mature with increasing cell organs,but also expressed the neuronal cell markers. This study showed that MSCs may represent an alternative stem cell source for neurotransplantation.
