摘    要:目的]研究不同型号大孔吸附树脂分离纯化生精汤中总黄酮的工艺条件。方法]以总黄酮含量为考察指标,通过比较4种大孔吸附树脂上的吸附与解吸附特性,筛选最佳树脂,并对该树脂的吸附特性及各影响因素进行系统研究,确定生精汤中总黄酮的最佳纯化工艺。结果]最佳纯化工艺为:样品液以1.0mL.min-1速度上样后在D101大孔吸附树脂静态吸附3.5h,先用5 BV水洗脱除杂,再用5 BV 70%乙醇解吸,洗脱流速为2.0mL.min-1,最大吸附量为16.21mg.g-1,所得产品中总黄酮平均含量为77.6%。结论]D101大孔吸附树脂对生精汤中总黄酮具有良好的吸附分离性能,吸附快、解吸附容易,性能优于其他树脂。

Abstract:Objective ]To study the optimal isolation and purification technologies of total flavones from Shengjing Tang(TFS) with different macroporous resins. Methods]Static and dynamic adsorption desorption methods were adopted to choose the optimal type of resin with the content of total flavones as the standard. The adsorption function of different macroporous resins and the effects of sample concentration, flow rate and eluant etc. were studied. Results ]The appropriate technological conditions were as follows: velocity of sample solution was 1.0 mL/min, 5 BV of water was used as purificant and 5BV of 70% ethanol was used as eluant,velocity of 70% ethanol was 2mL/min and the saturated adsorption capacity of D101 was 16.21mg/g. Under these conditions, the average content of total flavonoids extract was 77.6%. Conclusion] D101 resin is effective in the adsorption and purification of the total flavones from Shengjing Tang with good repetition and stability.
