人参有效单体对活化的肾成纤维细胞生长及TGF-β表达的影响

摘    要:[目的]观察人参有效单体对活化的肾成纤维细胞生长及TGF-β表达的影响。[方法]采用体外培养肾成纤维细胞的方法,将其分为空白对照组、TGF-β1 2ng·mL-1组、TGF-β1 2ng·mL-1+中药10-5g·mL-1组和TGF-β1 2ng·mL-1+中药10-6g·mL-1组。采用MTT法观察24h中药单体对肾成纤维细胞的作用,采用ELISA法检测其对TGF-β的分泌情况。[结果]与空白对照组相比,加入TGF-β1可以促进肾成纤维细胞的活化(P<0.05),以及TGF-β的分泌(P<0.01)。加入中药单体后人参三醇和人参皂苷Rf对活化的NRK-49F细胞抑制效果好(P<0.05),其次是人参皂苷Rg1和人参皂苷Rc(P<0.05)。人参皂苷Rd、人参皂苷F2、人参皂苷Rc和人参皂苷Rf对抑制活化的NRK-49F细胞TGF-β分泌效果好(P<0.05),其次是人参皂苷F1(P<0.05)。同种中药单体10-6 g·mL-1优于10-5 g·mL-1(P<0.05)。[结论]人参中药单体可以抑制活化的肾成纤维细胞的增殖,有效防治肾纤维化,其可能的作用机制与其抑制TGF-β的分泌有关。

The Effect of Utility Monomer in Ginseng to the Growth and Transforming Growth Factor-J5 Expression of Renal Fibroblast Cell

Abstract:[ Objective ] To observe the effect of utility monomer in ginseng to the renal fibroblast cell production and TGF-β expression. [Methods] To culture in vitro the renal fibroblast cell, which were divided into blank control group, TGF-β1 2ng·mL^-1 group, TGF-β1 2ng.mL^-1 + Chinese medicine monomer 10^-5 g·mL^-1 group and TGF-β1 2ng·mL^-1 + Chinese medicine monomer 10^-6g·mL^-1 group. To observe the effect of Chinese medicine monomer for 24 hours on the cell by MTT assay, and the secretion of TGF-β by ELISA assay. [Results]Compared with blank control group, adding TGF-β1-stimulating factor can promote the cell, as well as the secretion of TGF-β. Adding to the traditional Chinese medicine monomer, panaxatriol and ginsenoside Rac can inhibit the activation of the cell proliferation (P 〈0.05 ), followed with ginsenoside Rgl and ginsenoside Rc (P 〈 0.05). Ginsenoside Rd, ginsenoside F2 ginsenoside Rc and ginsenoside Rf can inhibit the secretion of TGF-β, followed ginsenoside F1 (P 〈0.05). The same kinds of traditional Chinese medicine monomer 10^-6g· mL^-1 was better than 10^-5 g.mL^-1 [Conclusion] Utility monomer of ginseng can inhibit the activation of the renal fibroblast cell proliferation and secretion of TGF-β, effective prevention and treatment of renal fibrosis, and its possible mechanism of action is related to inhibition of TGF-β secretion.
