摘    要:目的 验证宣痹通瘀胶囊对实验性大鼠血液流变学、体内动脉血栓及体外动-静脉旁路血栓形成的影响。方法 将大鼠随机分为空白组、血瘀模型组、通心络胶囊对照组、宣痹通瘀胶囊高、中、低剂量组。各实验组采用不同浓度、相同体积的方法灌胃给药。采用复合致瘀因素复制大鼠血瘀证动物模型。血液流变仪检测不同切变率的全血黏度及血浆黏度;采用电刺激大鼠颈总动脉导致局部血栓形成的方法,观察各组动物局部动脉血栓形成的时间;采用大鼠体外动-静脉旁路血栓形成的方法,检测各组动物体外血栓的湿重和干重。结果 与模型组比较,高、中、低剂量组不同切变率的全血黏度及血浆黏度均明显降低(P < 0.01或P < 0.05);高、中剂量组体内血栓形成时间均明显增加(P < 0.05);高剂量组形成的体外血栓湿重和干重均明显降低(P <0.05);中、低剂量组形成的体外血栓干重明显降低(P < 0.05)。结论 宣痹通瘀胶囊的预防性给药,对大鼠血瘀证的血液流变学特征具有明显改善作用,具有降低总血栓风险的作用,其治疗效果与用药剂量呈正相关。

Abstract:Objective To verify the effects of Xuanbi Tongyu Capsule on hemorheology, arterial thrombosis in vivo and arteriovenous bypass thrombosis in vitro in rats. Methods Rats were randomly divided into blank group, blood stasis model group, Tongxinluo capsule control group, Xuanbi Tongyu capsule high, medium and low dose groups. Each experimental group was administered by gavage with different concentration and the same volume. The animal model of blood stasis syndrome in rats was duplicated by compound factors of blood stasis. The whole blood and plasma viscosities at different shear rates were measured by hemorheology instrument; the time of local arterial thrombosis was observed by stimulating common carotid artery in rats; the wet and dry weight of thrombosis in rats were measured by arteriovenous bypass thrombosis in vitro. Results Compared with model group, the whole blood viscosity and plasma viscosity at different shear rates in high, medium and low dose groups were significantly decreased (P < 0.01 or P < 0.05), the time of thrombosis in high and medium dose groups was significantly increased (P < 0.05), and the wet and dry weight of thrombus in vitro in high dose groups were significantly decreased (P < 0.05). The dry weight of thrombus in vitro in the dose group decreased significantly (P < 0.05). Conclusion : Preventive administration of Xuanbi Tongyu Capsule can significantly improve the hemorheological characteristics of blood stasis syndrome in rats and reduce the risk of total thrombosis. The therapeutic effect is positively correlated with the dosage of Xuanbi Tongyu Capsule.
