摘    要:目的 建立快速、灵敏、高效的精氨酸双糖苷的测定方法并研究精氨酸双糖苷在大鼠体内的药动学。方法 以乌苯美司作为内标,采用LC-MS/MS检测方法测定血浆中精氨酸双糖苷浓度,利用DAS2.0软件计算药动学相关参数,并分析方法的可靠性。结果 空白血浆对检测物质不产生干扰,在20~20 000 ng/mL符合线性关系,方法学中RSD以及RE值均小于15%。结论 采用LC-MS/MS方法检测精氨酸双糖苷方法合理可靠,精氨酸双糖苷的绝对生物利用度为(4.50±3.75)%。

Abstract:Objective To develop a quick, specific and high performance method for determination of Arginyl-fructosyl-glucose (AFG) and to investigate pharmacokinetics of AFG in rats in vivo. Methods Bestatin is used as an internal standard, the concentration of AFG in plasma was determined by LC-MS/MS. The DAS 2.0 software was adopted to calculate bioavailability of AFG. Results Blank plasma does not interfere with the test substance, there is a significant correlation in the range of 20 ng/mL to 20 000 ng/mL, and the RSD and RE values are less than 15% in the method. Conclusion The average relative bioavailability of AFG was (4.50 ±3.75)%, The measured method of AFG by LC-MS/MS is reasonable and reliable.
