新生儿缺氧缺血性脑病脑细胞凋亡研究

摘    要:目的 探讨因患缺氧缺血性脑病(hypoxlc ischemic cnccphalopath,HIE)死亡新生儿的脑组织是否存在细胞凋亡情况。方法 收集因HIE死亡新生儿5例(HIE组)及非因HIE死亡新生儿2例(非HIE组),以肺活检穿刺针自前囟穿刺取脑组织,通过HE染色、原位末端脱氧核苷酸转移酶标记法(TUNEL)及电镜等方法,观察脑细胞病理形态变化,计数单位面积下凋亡阳性细胞数。结果 HIE组新生儿脑细胞在光镜及电镜下观察可见较多凋亡细胞及坏死细胞,TUNEL,染色凋亡细胞阳性率平均约10.90个/mm^2,非HIE组新生儿脑细胞少见凋亡,阳性率约3.50个/mm^2,电镜下细胞结构保存完整。结论 患HIE死亡新生儿脑细胞存在凋亡,从在体研究方面证实细胞凋亡参与新生儿HIE发病机制。

Abstract:Objective To explore whether or not apoptosis occurs in neonatal infants with hypoxic ischemic encephalopathy(HIE).Methods 5 neonates died of HIE,which were HIE group,and 2 neonates died not of HIE,which were non HIE group,were collected,whose cortices were taken using a lung biopsy paracentetic needle.Using HE staining,terminal deoxynuleotidyl transferase mediated dUTP nick end labeling(TUNEL) and electronmicroscope technique,the pathologic features of the cells were observed and the positive numbers of the apoptosis cells in unite area were counted.Results The apoptosis and necrosis cells could obviously be observed from the HIE group under light and electron microscope,however they were seldom seen from the non HIE group.The average positive cells in unit area were 10.90 pc.per mm 2 and 3.50 pc.per mm 2 respectively.Conclusion Cell apoptosis occurred in neonates died of HIE,confirming that the apoptosis have taken part in the pathogenesis of the HIE in vivo.
