摘    要:目的探讨前列腺干细胞抗原(PSCA)在人子宫内膜样腺癌内膜组织中的表达及与临床病理学特征的关系。方法采用免疫组化、荧光定量RT—PCR和Western Blot方法检测子宫内膜腺上皮细胞中PSCA的表达。结果PSCA在内膜样腺癌组织主要表达于癌细胞,细胞间质和肌肉组织均无表达。从单纯性、复杂性到不典型增生,PSCA在mRNA、蛋白水平上的表达评分均有逐渐增多趋势,不典型增生与前二者相比,差异均有统计学意义(P〈0.01)。在内膜样腺癌中,随着分化程度的降低、分期的增加和浸润深度的增加,PSCA mRNA、蛋白表达逐渐增加。在增生期和分泌期PSCA的表达差异无统计学意义。PSCA表达水平与临床分期、病理分级和浸润深度均有相关性(P〈0.05)。结论PSCA是一个新的细胞表面抗原,可能参与了癌前病变的发生,其表达水平在一定程度上反映了内膜样腺癌的生物学行为,并介导了内膜样腺癌的发生与发展,可能在内膜样腺癌诊断、免疫治疗等方面具有广阔的应用前景。

Abstract:Objective To analyze the expression of prostate stem cell antigen (PSCA) in gland epithelial cells of various endometriums. Methods The Immunohistochemistry, RT-PCR and Western Blot method were used to detect the PSCA expression in various endometriums. Results PSCA in endometrioid adenocarcinoma was mainly expressed in cancer cells, stromal cells, and no expression in muscle tissue. PSCA become increasing from Simple Hyperplasia (SH), Complex Hyperplasia(CH) and Atypical Hyperplasia (AH),particularly in AH (P<0.01). In the Endometrioid Adenocarcinoma (EAC), PSCA expression was associated with the differentiation grades (DG), myometrial invasion and surgical pathological stages. Poorer of DG, more deeper of myometrial invasion, and later of more of surgical pathological stages, more PSCA expression. The expression of PSCA was no significant difference between the proliferative phase and secretory phase. RT-PCR and Western Blot have got the same results with IHC. Conclusion PSCA may participate in the origin of precancerous lesions. PSCA expression may related to the bio-logical behavior of EAC and the development of EAC.
