白花蛇舌草对正常器官氧化损伤防护效应的实验研究

摘    要:目的探讨白花蛇舌草对经抗肿瘤药物治疗的荷瘤小鼠正常器官氧化损伤的防护效应及机制。方法荷瘤鼠随机分为3组:对照组,不给予任何治疗继续饲养至实验结束;单纯化疗组,单次腹腔注射顺铂(DDP)或阿霉素(ADM);化疗+白花蛇舌草组,单次腹腔注射DDP或ADM,并给予白花蛇舌草水煎液灌胃,连续应用7天。化学比色法测定荷瘤小鼠肝脏、脾脏、肾脏和骨髓组织的活性氧(ROS)、丙二醛(MDA)水平和谷胱甘肽过氧化酶(GSH-Px)、过氧化氢酶(CAT)活性,电镜下观察脾脏超微结构变化。结果化疗+白花蛇舌草组小鼠肝、脾和骨髓组织的ROS、MDA水平和GSH-Px、CAT活性较单纯化疗组明显降低(P〈0.01),脾脏超微结构变化明显减轻。结论白花蛇舌草具有抗氧化损伤作用,可减轻化疗药物引起的荷瘤鼠正常器官的副损伤。

Abstract:Objective To study the effect of Hedyotic diffusa on the oxidative injury of the normal tissues induced by antitumorous drugs in tumor-bearing mice.Methods The contents of reactive oxygen species(ROS) and malondialdehyde(MDA) and the activities of glutathione peroxidase(GSH-Px) and catalase(CAT) in tissue homogenates were determined by chemocolorimetry.Electron microscope was used to study the ultrastructure of spleen.Results Hedyotic diffusa reduced the activities of GSH-Px and CAT and reduced the contents of ROS and MDA in the normal tissues of tumor-bearing mice treated with chemotherapy.Hedyotic diffusa also lessened the ultrastructural injury of spleen.Conclusion Hedyotic diffusa has protective effects against oxidative injury of normal tissues induced by antitumorous drugs in tumor-bearing mice.
