COPD大鼠膈肌组织泛素-蛋白酶体途径的研究

摘    要:目的研究COPD大鼠膈肌组织内蛋白降解途径之一的泛素-蛋白酶体系统组成成分的变化及意义。方法采用单纯熏香烟法复制COPD大鼠动物模型。蛋白免疫印记(Westernblot)方法测定大鼠膈肌内E2-14k、蛋白酶体C8亚基的蛋白表达;逆转录-聚合酶链式反应(RT-PCR)技术检测膈肌内泛素和蛋白酶体C2亚基的mRNA表达。结果Westernblot分析结果显示,COPD大鼠膈肌组织中E2-14k的蛋白表达较正常对照组显著增强(0.81±0.28比0.50±0.25,P〈0.05),蛋白酶体C8亚基的蛋白表达变化不明显(P〉0.05);RT-PCR分析结果显示,膈肌组织中泛素的mRNA表达(IOD值)较对照组显著增加(0.89±0.20比0.50±0.15,P〈0.05),蛋白酶体C2亚基的mRNA表达与正常对照组比较无明显差异(P〉0.05)。结论COPD大鼠膈肌组织内泛素-蛋白酶体降解途径被激活,可能是引起蛋白降解的重要因素之一。
