人白细胞介素18对肺癌A549细胞生物学功能的影响

摘    要:目的 探讨人白细胞介素18(interleukin 18,IL-18)对肺癌A549细胞生物学功能的影响。方法 将IL-18重组质粒及其空载对照质粒转染肺癌A549细胞,用抗生素加压筛选阳性细胞,构建稳定表达IL-18质粒及空载质粒的肺癌细胞,将构建好的稳定过表达IL-18细胞和空载对照细胞分别命名为IL18-A549细胞和NC-A549细胞。采用细胞计数法、流式细胞术、划痕实验和transwell迁移、侵袭实验检测IL-18过表达对肺癌A549细胞增殖、凋亡、运动、迁移和侵袭能力的影响。结果 成功构建稳定过表达IL-18肺癌细胞IL18-A549。与空载对照细胞NC-A549相比,过表达IL-18可促进肺癌A549细胞增殖(P<0.05),但对细胞早期、晚期及总凋亡率无明显影响(P>0.05)。IL18-A549细胞的划痕愈合率为(62.0±5.7)%,高于NC-A549细胞的(31.2±3.7)%,两者比较差异有统计学意义(P<0.05);IL18-A549细胞和NC-A549细胞发生迁移的细胞数目平均为48个和9个,发生侵袭的细胞数目平均为33个和11个,两者差异均有统计学意义(P均<0.05)。结论 过表达IL-18可促进肺癌A549细胞增殖、运动、迁移和侵袭,IL-18可能是肺癌转移过程中的关键因子。

Abstract:Objective To investigate the biological functions of IL-18 on the human lung cancer cell A549,including its role in cancer-related processes. Methods A plasmid expressing IL-18 or a control plasmid were transfected into the A549 human lung cancer cell line,and antibiotic selection was used to identify stable transfectants. The resulting respective cell lines,IL18-A549 and NC-A549,were compared in terms of proliferation, apoptosis, movement,invasion and migration. Results The IL18-A549 cell line stably expressed IL-18, and comparison with the NC-A549 cell line suggested that IL-18 promotes proliferation(P<0.05). The rate of healing in a wound healing assay was significantly higher for IL18-A549 cells(62.0±5.7)% than for control cells (31.2±3.7)%(P<0.05),and the percentages of cells showing migration and invasion in a transwell assay were significantly higher for IL18-A549(48% and 33%) than for controls (9% and 11%;P<0.05). Flow cytometry showed similar rates of early,late and total apoptosis between IL-18 transfectants and control cells. Conclusion IL-18 promotes proliferation,movement,invasion and migra-tion of A549 cells,and may play a key role in human lung cancer. Inhibiting IL-18 function may be useful for treating lung cancer.
