摘    要:目的:研究阿坎酸对大鼠局灶性脑缺血-再灌注损伤的神经保护作用.方法:大鼠大脑中动脉栓塞模型参照Longa报道的方法进行.48只SD雄性大鼠随机分为假手术组、模型组、阿坎酸组.缺血2 h后恢复灌注,给药组腹腔注射给药.造模前后进行神经损害严重程度评分(NSS),TTC染色测定梗死面积,HE染色检测脑组织病理变化,应用蛋白质印迹检测大鼠脑组织中NR2B的表达.结果:阿坎酸能明显改善大鼠的神经行为,缩小梗死面积,并使脑组织病理改变减轻.同时,阿坎酸能有效降低NR2B的表达.结论:阿坎酸对局灶性脑缺血/再灌注大鼠有明显的保护作用.

Abstract:OBJECTIVE To investigate the neuroprotective effects of acamprosate on cerebral ischemia reperfusion (CIR) injury in rats. METHODS Rat middle cerebral artery embolism model referred to Longa reported methods. Forty eight male SD rats were randomly divided into control group, model group and acamprosate group. After reperfusion for 22 h, the neurologic deficit score and infarction area were measured. Then pathological change in injury brain tissues was detected by HE staining. The expression of NR2B was measured using western blot. RESULTS Acamprosate significantly improved rat neural behaviors, reduced cerebral infarction area, relieved injuries of brain tissues, and reduced expression of NR2B.CONCLUSION Acamprosate has obvious neuroprotective effects on cerebral ischemia/reperfusion injury in rats.
