miR-145和miR-497在宫颈癌SiHa细胞中的表达及对增殖的影响

摘    要:
目的 研究miR-145和miR-497在宫颈癌SiHa细胞的表达及对增殖的影响。方法 以宫颈癌SiHa细胞为研究对象,转染pre-miR-145和pre-miR-497,采用实时荧光定量PCR和MTT检测miR-145和miR-497在宫颈癌SiHa细胞中的表达及对增殖的影响。结果 采用实时荧光定量PCR检测miR-145和miR-497在宫颈癌SiHa细胞中的表达,结果显示,实验组中miR-145和miR-497的表达量显著高于blank组(P=0.001),MTT结果显示转染miR-145和miR-497的SiHa细胞其增殖活性明显受到抑制(P<0.05)。结论 miR-145和miR-497能够显著抑制宫颈癌SiHa细胞的增殖。

Abstract:
Objective To evaluate the effect of miR-145 and miR-497 on proliferation of human cervical carcinoma cells and their expression.Methods Pre-miR-145 and pre-miR-497 were transfected in cervical cancer SiHa cell,and detected their expression and effect on proliferation using real-time fluorescent quantitative PCR and MTT.Results The Results showed that the experimental group in the expression of miR-145 and miR-497 amount was significantly higher than that of blank group(P=0.001),and MTT showed that miR-145 and miR-497 can significantly suppressed the proliferation of SiHa(P<0.05).Conclusion miR-145 and miR-497 could inhibit the proliferation of SiHa cells.
