三氧化二砷诱导骨肉瘤U2OS细胞凋亡的作用

 目的 探讨三氧化二砷(arsenic trioxide, AS₂O₃)诱导骨肉瘤U2OS细胞凋亡的作用。方法 将AS₂O₃按不同浓度(0、4、8 μmol/L)作用于细胞24 h后,分别通过光镜、MTT法、Hochst33342染色检测细胞的凋亡情况,另以8 μmol/L的AS₂O₃作用细胞,通过流式细胞仪分别在8、16、24 h后,观察细胞凋亡情况。结果 AS₂O₃可抑制骨肉瘤U2OS细胞的增殖,浓度越大,抑制越强,4 μmol/L的AS₂O₃作用细胞24 h后,增殖率为(66.32±5.15)%,8 μmol/L的AS₂O₃作用细胞24 h后,增殖率为(26.73±9.36)%,并且作用时间越长,抑制作用越强,8 μmol/L的AS₂O₃作用于U2OS细胞8、16、24 h后,凋亡率分别为21.16%,36.15%,47.13%。结论 AS₂O₃可诱导骨肉瘤U2OS细胞的凋亡,抑制肿瘤的增殖。

A study of arsenic trioxide to induce apoptosis of osteosarcoma U2OS cells

Objective To study the effect of arsenic trioxide on inducing apoptosis of osteosarcoma U2OS cells in vitro. Methods Osteosarcoma U2OS cells were treated with arsenic trioxide at different concentrations(0,4 μmol/L,8 μmol/L) for 24 h, respectively, then were observed under microscope,by MTT assay and Hochst33342 staining method. U2OS cells were treated with arsenic trioxide at 8 μmol/L and then detected by flow cytometry at 8 h, 16 h and 24 h. Results AS₂O₃ could inhibit U2OS cells growth, the U2OS cells proliferation rate was (66.32±5.15)% after treatment with arsenic trioxide at 4umol/L concentration, and (26.73±9.36)% at 8 μmol/L concentration. The U2OS cells apoptosis rate was 21.16%,36.15%,47.13%respectively at 8 h, 16 h, 24 h after treatment with AS₂O₃. The effect of arsenic trioxide on inhibiting U2OS cells growth is dependent on time and concentration. Conclusions AS₂O₃ can induce apoptosis of osteosarcoma U2OS cells and inhibit the cells growth.
