超高效液相色谱法测定肝病患者氟康唑血药浓度

目的:建立用超高效液相色谱法测定肝病患者血中氟康唑的浓度。方法:采用超高效液相色谱(UPLC) 法,Zorbax Eclipse plus-C₁₈柱(2.1 mm×100 mm,1.8 μm),保护柱Zorbax SB-C₁₈柱(2.1 mm×5 mm,1.8 μm),以乙腈-水(24:76)为流动相,在210 nm 处测定其含量。结果:线性范围为0.1~20 μg·ml^-1,样品提取回收率为80.95%(n=5),方法回收率为98.67%~106.37%,日内精密度RSD≤4.21%,日间RSD≤2.17%,且稳定性良好。结论:该方法可靠、简便、快速,可为临床合理用药提供参考。

Plasma concentration of fluconazole in patients with liver disease determined by ultra performance liquid chromatography

OBJECTIVE To determine plasma concentration of fluconazole in patients with liver disease by ultra performance liquid chromatography (UPLC). METHODS Plasma fluconazole concentration in patients with liver disease was determined by UPLC. Analytical columns: Zorbax Eclipse plus-C₁₈(2.1 mm×100 mm, 1.8 μm), protecting columns: Zorbax SB-C₁₈ (2.1 mm×5 mm, 1.8 μm); mobile phase: acetonitrile-water (24:76); detection wave length: 210 nm.RESULTS Linear range was 0.1-20 μg·ml^-1. Intra-day and inter-day precisions were less than 4.21% and 2.17%, respectively. Extraction recovery of fluconazole was 80.95% (n=5), and method recoveries were 98.67%-106.37%. CONCLUSION The method is reliable, convenient and efficient, which can provide a reference for clinical rational drug use.
