摘    要:
目的:建立民族药铁包金提取物中黄酮类成分含量测定的一测多评法,探讨一测多评法在民族药质量控制中的应用。方法:以儿茶素为内参物,分别建立花旗松素、香橙素、圣草酚和槲皮素与儿茶素的相对校正因子,分别采用外标法和一测多评法计算测定5种黄酮的含量,并比较二者结果相对误差。结果:4种黄酮类成分相对于儿茶素的相对校正因子分别为fcatechin/Taxifoli280 nm=1.229,fcatechin/Aromadendrin280 nm=1.145,fcatechin/Eriodictyol280 nm=1.364,fcatechin/Quercetin280 nm=0.897;相对保留时间分别为RTaxifoli/Catechin=1.49,RAroma dendrin/Catechin=2.22,REridictyol/Catechin=3.30,RQuercetin/Catechin=3.72;一测多评法的计算结果与外标法的实测值一致,实验所得的相对校正因子可靠、相对保留时间稳定。结论:建立的一测多评法可作为铁包金提取物中黄酮类成分的含量测定方法,一测多评法为民族药质量控制提供了一个新的模式与方法。

Abstract:
OBJECTIVE To simultaneously determine contents of five flavones in ethnic medicine Berchemia lineata extract by quantitative analysis of multiple components with a single-marker (QAMS), and discuss application of QAMS in quality control of ethnic medicines. METHODS Catechin was chosen as internal reference substance. Relative correlation factors (RCF) of taxifolin, aromadendrin, eriodictyol and quercetin to catechin were calculated and established. Contents of these five flavones were determined by external standard method (ESM) and QAMS, respectively, and relative error (RE) between QAMS and ESM was analyzed to evaluate QAMS method. RESULTS Within linear ranges, values of RCF determined at 280 nm were f280 nmcatechin/Taxifoli=1.229,f280 nmcatechin/Aromadendrin=1.145,f280 nmcatechin/Eriodictyol=1.364,f280 nmcatechin/Quercetin=0.897,respectively. RCF showed good reproducibility with a RSD<3%. Relative retention times (RRT) were RTaxifoli/Catechin=1.49,RAroma dendrin/Catechin=2.22,REridictyol/Catechin=3.30 and RQuercetin/Catechin=3.72. RRT showed good stability with a RSD<5%. Two methods did not show significant difference in assay results. CONCLUSION QAMS method is feasible, credible, and can be used to determine multiple flavones in Berchemia lineata extract. QAMS can be adopted as a novel strategy for quality control of ethnic medicines.
