高效液相色谱法同时测定丹参白疕消丸中6种有效成分的含量

目的:建立丹参白疕消丸同时测定盐酸小檗碱、丹皮酚、丹参酮ⅡA、大黄素、大黄酚和大黄素甲醚6种成分含量的HPLC 测定方法.方法:采用高效液相色谱法,Inertsil ODS-3色谱柱(250 mm×4.6 mm,5 μm),柱温25 ℃,流动相:甲醇-0.4%磷酸水溶液梯度洗脱,流速0.8 ml·min^-1,检测波长为354 nm(盐酸小檗碱)、274 nm(丹皮酚)、254 nm(大黄酚、大黄素、大黄素甲醚和丹参酮ⅡA),外标法计算含量.结果:盐酸小檗碱、丹皮酚、大黄素、丹参酮ⅡA、大黄酚和大黄素甲醚6种成分分别在0.1939~1.5515,0.0417~0.3333,0.0625~0.5000,0.0784~0.6272,0.2120~1.6963,0.3584~1.0754 μg与峰面积线性关系良好(r²> 0.9982),样品平均回收率为95.67%~98.14%,RSD< 1.85%.结论:该方法快速、简便、灵敏、重复性好,可用于丹参白疕消丸的质量控制方法.

Simultaneous determination of six components in Danshenbaibixiao Pills by HPLC-DAD

OBJECTIVE To establish a method for simultaneous determination of berberine hydrochloride, paeonol, emodin, tanshinoneⅡA, chrysophanol and physcion in Danshenbaibixiao pills by HPLC. METHODS Separation was performed on an Inertsil ODS-3 column (150 mm×4.6 mm,5 μm) of HPLC. The mobile phase consisted of methanol(A)-0.4% phosphoric acid (B) water (51:49) with gradient elution at 25 ℃. The flow rate was 0.8 ml·min^-1 and at a detection wavelength of 354 nm (berberine hydrochloride), 274 nm (paeonol), 254nm (emodin, tanshinoneⅡA, chrysophanol, physcion). The determination was a external standard method. RESULTS Calibration curves of all active constituents showed good linearity (r² >0.9982) in a relatively wide concentration range. The liner range of the active constituents was 0.1939-1.5515,0.0417-0.3333,0.0625-0.5000,0.0784-0.6272,0.2120-1.6963 and 0.3584-1.0754 μg,respectively. The average recovery was 95.67-98.14%, RSD <1.85%. CONCLUSION This method is fast, simple, sensitive, and repeatable, and can be used for determination of components in Danshenbaibixiao pills.
