2016不同浓度稀释碘伏联合顺铂对SGC-7901胃癌细胞生长的影响

目的 进一步探讨不同浓度稀释碘伏对体外培养SGC-7901细胞生长的影响。方法 体外培养SGC-7901细胞,待其生长至对数期时,接种96孔板,至细胞贴壁后分别以浓度梯度的稀释碘伏及碘伏与顺铂联合处理SGC-7901细胞,MTT法检测两种方案的药物对细胞增殖的影响;AO/EB染色方法 观察其对肿瘤细胞形态学改变的影响。结果 浓度梯度的稀释碘伏及碘伏联合顺铂对SGC-7901细胞的生长均有明显的增殖抑制作用,且呈浓度时间依赖性;AO/EB染色结果可见:随药物浓度的增大,活细胞量逐渐减少,胞质变少,体积缩小、胞核浓缩,于镜下可见凋亡小体。结论 稀释碘伏对SGC-7901细胞的增殖抑制作用显著,对细胞凋亡具有一定的诱导作用。

Effect of povidone-iodione on growth of human gastric cancer cell line SGC-7901

Objective To explore the effects of diluted povidone-iodione on human gastric cancer cell line SGC-7901 growth in vitro.Methods Gastric cancer cell line SGC-7901 was inoculated in a 96 well cell culture plate when grew to the logarithmic phase.Then SGC-7901 cells were treated with concentration gradient of diluted povidone-iodione and the mixture of diluted povidone iodine and cisplatin,respectively.MTT assay was used to examine the proliferation of the two schemes.AO/EB staining method was used to observe its effect on the morphological changes of tumor cells.Results The diluted povidone-iodione and the mixture of diluted povidone-iodine and cisplatin showed obvious proliferation inhibition effect on SGC-7901 cells,with concentration- and time- dependence effect.AO/EB staining Results showed that, with the increase of drug concentration,the amount of living cells gradually reduced,cytoplasm became less,cell volume decreased,nucleus concentrated and apoptotic bodies could be seen under the microscope.Conclusion Diluted povidone-iodione can significantly inhibit the proliferation of SGC-7901 and induce the apoptosis of SGC-7901 in a certain extent.
